Mithilfe kryo-elektronenmikroskopischer Bilder in bisher unerreichter Auflösung konnte Wilson mit seinem Team nun neue Einblicke in die Resistenzbildung gegen das Antibiotikum Erythromycin gewinnen, wie die Wissenschaftler im Fachjournal Molecular Cell berichten.
“Ein besseres Verständnis dieser Mechanismen ist ein wichtiger Schritt auf dem Weg, neue, wirksame Antibiotika zu entwickeln”, sagt Wilson.

As they report in the journal Molecular Cell, he and his team have used high-resolution cryo-electron microscopy to obtain novel insights into the ultrastructural changes in an intracellular machine associated with the acquisition of resistance to the antibiotic erythromycin.
“A better understanding of this mechanism is an important step towards the development of new and more potent antibiotics,” Wilson says.

Stattdessen scheint die Anwesenheit von Erythromycin die Eiweißkette, aus der das Signalpeptid besteht, im Inneren des Ribosoms umzuleiten. Dabei nimmt ErmBL eine spezielle Struktur an, die das aktive Zentrum des Ribosoms hemmt.
„Diese Erkenntnisse könnten zukünftig helfen, bessere Makrolidantibiotika zu entwickeln“, ist Wilson überzeugt.
