In skeletal muscle cells of the zebrafish, Linke’s team explored what happens when the protection by the methylated heat shock protein is repressed.
By genetic manipulation they altered the organism in such a way that it no longer produced the enzyme Smyd2, which blocked the methylation of Hsp90.

Verantwortlich für diese Modifikationen sind diverse Enzyme und Proteine, die in variablen Komplexen die DNA methylieren, an Histonen verschiedene chemische Gruppen, u.a. Methylreste, koppeln oder als Transkriptionsfaktoren an entsprechend umstrukturiertem Chromatin die Transkription spezifischer Gene regulieren.
Unser Forschungsanliegen beinhaltet die Histondemethylasen wie KDM1A, die gerade in embryonalen Tumoren überexprimiert und in hohem Maße an der Regulation von zellulären Proliferations-, Differenzierungs- und Migrationsprozessen beteiligt sind.

Various enzymes and proteins, mostly part of variable complexes, are responsible for these modifications. They methylate DNA and couple different chemical groups to histones, including methyl residues. Additionally, transcription factors regulate transcription of specific genes in the accordingly reorganized chromatin.
Our research is including the analyses of histone demethylases, especially KDM1A that are overexpressed in embryonic tumors and currently highly involved in the regulation of cellular proliferation, differentiation and migration processes.

Es kommt zu einer globalen Hypomethylierung, d.h. insgesamt ist die DNA geringer methyliert als im gesundem Gewebe und zu einer Hypermethylierung, einzelner Gene, vor allem von Tumorsupressorgenen.
Durch die verstärkte DNA-Methylierung werden Tumorsupressorgene, welche normalerweise die Entstehung von Tumoren verhindern, in Tumoren geringer exprimiert als im gesunden Gewebe.

a global hypomethylation (overall the DNA is less methylated compared to healthy tissue) and a hypermethylation of specific genes, in particular of tumor suppressor genes, occurs.
As a consequence of the increase in DNA-methylation the tumor suppressor genes are less active (expressed) in tumor tissues compared to healthy tissues.
